In order to understand the mechanism and the regulation of the synthesis of the histone H3.2 variant, we established a model system using mouse embryo fibroblast C3H10T1/2 cells which are an excellent tool for studying the regulation of cell cycle progression. In this report, we describe the development of an in vitro model system of H3.2 synthesis. In undifferentiated C3H10T1/2 cells grown for 24 h in Dulbecco’s modified Eagle’s medium supplemented with 10% fetal bovine serum, the H3 variant was not detectable by western blot analysis. In C3H10T1/2 cells grown to confluency in 100% FCS, H3.2 was synthesized, as revealed by the presence of a single band of the expected molecular weight (Mr = 15 kDa) by western blot analysis. C3H10T1/2 cells can be adapted in vitro to achieve a complete commitment to differentiation. Under these conditions, cells stop proliferating and start to differentiate. The differentiated cell-type expresses specific markers of endodermal, mesodermal, ectodermal and neural tissues. Within the endodermal cell-type, we have shown the synthesis of H3.2 variant to be completely downregulated. These results therefore demonstrate that the in vitro system developed in this study is a useful tool for studying the regulation of histone H3.2 synthesis. Furthermore, they also demonstrate that, in a transcription- and translation-independent manner, H3.2 synthesis is under the control of a differentiation process.All for one and one for all?
