A total of 38 isolates of enteropathogenic Escherichia coli (EPEC) were obtained from children hospitalized with diarrhea in a Pediatric Hospital in Madrid, Spain, between January 1999 and December 2001. Using serotyping, 20 different O serogroups were identified. All belonged to serogroup O27, and were assigned to the different typing groups F3 (n = 27), E2 (n = 4), and G3 (n = 5). More than 85% of isolates possessed the structural and regulatory genes of the locus encoding the phospho-N-acetylmuramoyl-L-alanine amidase (P muropeptidase) of the LEE pathogenicity island. The remaining isolates harbored the genetic determinants that encode protein A (prtA), the enterobacterial common antigen, and the locus that encodes the translocated intimin (tir). The intimin and translocated intimin receptor genes were amplified by polymerase chain reaction, and the products were sequenced. The intimin gene detected had been described previously and was designated intimin eae. The novel truncated intimin tvi was classified as intimin V. The intimin V and intimin eae genes were closely related, sharing 85% similarity at the nucleotide level. The tir gene was not present in these isolates, suggesting that these EPEC strains may not encode a functional intimin receptor.
