The objective of this study was to investigate the role of microRNAs in the regulation of NICD1 expression in ox-LDL-stimulated vascular smooth muscle cells (VSMCs). VSMCs were stimulated with ox-LDL and changes in the expression levels of miR-31, -32, and -146a-5p were monitored. Expression levels of miR-31, -32, and -146a-5p were determined in 20 premature aging patients (iPS) and 20 healthy donors (NH). Expression levels of P21, NICD1, and Jagged1 were also assessed in iPS and NH. In VSMCs, we demonstrate that ox-LDL induces cell cycle arrest and p21 expression via activation of the Notch-1 signaling pathway. Ox-LDL-mediated cell cycle arrest is associated with increased miR-31, -32, and -146a-5p expression. Flow cytometric analysis and western blotting also show that ox-LDL-induced p21 expression is attenuated by lentiviral-mediated miR-31 and miR-32 overexpression. Dual luciferase reporter assay demonstrates that Notch-1 is a direct target of miR-31, -32, and -146a
